Continuous flow Aza-Michael reaction for preparing the fast-acting synthetic opioid drug Remifentanil

چکیده
Remifentanil is a modern fentanyl analogue with ultrashort-action granted by an esterase-labile methyl propanoate chain. Here, we present the development of continuous flow methodology for key N-alkylation step remifentanil preparation in biphasic, “slug-flow” regime. We screened parameters under microwave-assisted reactions, translated conditions to settings, and obtained 15-min residence time 1-mL microreactor, space-time yield 89 mg/mL·h 94% yield.

Squaramide-catalysed asymmetric cascade aza-Michael/Michael addition reaction for the synthesis of chiral trisubstituted pyrrolidines.
A bifunctional squaramide catalysed aza-Michael/Michael cascade reaction between nitroalkenes and tosylaminomethyl enones or enoates has been developed. This organocatalytic cascade reaction provides easy access to highly functionalized chiral pyrrolidines with a broad substrate scope, giving the desired products in good yields (up to 99%) with good diastereoselectivities (up to 91 : 9 dr) and ...
متن کاملSynthesis, characterization and using Fe3O4@SiO2@FeCl3 as a new nanocatalyst for aza-Michael reaction between amines and ethyl crotonate
In this article, recyclable Fe3O4@SiO2@FeCl3 was synthesized and entirely characterized by various techniques including XRD, FT-IR, SEM, EDX and VSM analysis. The catalytic ability of produced Fe3O4@SiO2@FeCl3 was studied in the aza-Michael reaction of diethyl amine and ethyl crotonate which lead to high yie...
متن کاملMorphine deprivation increases self-administration of the fast- and short-acting mu-opioid receptor agonist remifentanil in the rat.
Opiate dependence and withdrawal have long been hypothesized to enhance the reinforcing effects of opiates; however, opiate agonist self-administration in these states has yet to be systematically assessed. To address this issue, the reinforcing property of the short-acting mu-opioid agonist, remifentanil, was assessed in morphine-dependent (MD), morphine-dependent and -withdrawn (MW), and nond...
